Trichostatin A
Trichostatin A (TSA) ist ein gegen Pilze wirksames Antibiotikum aus dem Bakterium Streptomyces platensis und blockiert selektiv die Klasse I- und Klasse II- Histon-Deacetylasen (HDAC) von Säugetieren; aber nicht HDAC der Klasse III/ Sirtuine.
TSA blockiert außerdem den eukaryontischen Zellzyklus während des Beginns der Zellteilung.
TSA kann genutzt werden, um die Genexpression zu beeinflussen, indem Acetylgruppen an Histonen entfernt werden und so Transkriptionsfaktoren Zugang zu zuvor verplombten Promotorsequenzen erhalten. Da in Krebszellen häufig Histon-Deacetylasen aktiv sind, die in Körperzellen abgeschaltete Gene aktivieren, besitzt das Trichostatin A Potential als Krebsmedikament. Eine Theorie zum Wirkmechanismus von TSA besagt, dass das Antibiotikum die Expression von Apoptose-Genen hochreguliert und so die Überlebenswahrscheinlichkeit von Krebszellen senkt. Andere Theorien sehen das TSA als Induktor der Zelldifferenzierung, was einen Wachstumsstop der ansonsten dedifferenzierten und somit teilungsfähigen Krebszellen bewirkt.
In klinischen Untersuchungen wird Trichostatin A bislang kaum eingesetzt.

HDAC inhibitors have been proposed as anticancer agents. However, their roles in innate genes expression remain not well known. Cathelicidin LL-37 is one of the few human bactericidal peptides, but the regulation of histone acetylation on LL-37 expression in airway epithelium remains largely unknown. Therefore, we investigated the effects of two non-selective HDACi, Trichostatin A (TSA) and sodium butyrate (SB), on the expression of the cathelicidin LL-37 in human airway epithelial cells. LL37 in human NCI-H292 airway epithelial cells and the primary cultures of normal nasal epithelial cells(PNEC) in response to HDAC inhibitors with or without poly (I:C) stimulation was assessed using real-time PCR and western blot. In parallel, IL-6 expression was evaluated by ELISA. Our results showed that HDAC inhibitors up-regulated LL-37 gene expression independent of poly (I:C) stimulation in PNEC as well as in NCI-H292 cells. HDAC inhibitors increased LL37 protein expression in NCI-H292 cells but not in PNEC. In addition, HDAC inhibitors significantly inhibited poly (I:C)-induced IL-6 production in both of the epithelial cells. In conclusion, HDAC inhibitors directly up-regulated LL-37 gene expression in human airway epithelial cells.

Cyclin D1 is an important regulator of G1-S phase cell cycle transition and has been shown to be important for breast cancer development. GSK3β phosphorylates cyclin D1 on Thr-286, resulting in enhanced ubiquitylation, nuclear export and degradation of the cyclin in the cytoplasm. Recent findings suggest that the development of small-molecule cyclin D1 ablative agents is of clinical relevance. We have previously shown that the histone deacetylase inhibitor Trichostatin A (TSA) induces the rapid ubiquitin-dependent degradation of cyclin D1 in MCF-7 breast cancer cells prior to repression of cyclin D1 gene (CCND1) transcription. TSA treatment also resulted in accumulation of polyubiquitylated GFP-cyclin D1 species and reduced levels of the recombinant protein within the nucleus.
